ERG344 inhibits the enzyme Aconitate Decarboxylase 1 (ACOD1), also known as Immune Responsive Gene 1 (IRG1). ACOD1 is known to catalyze the conversion of cis-aconitate to itaconate in the TCA cycle is of pro-inflammatory macrophages. The functions of ACOD1 appear to be context-dependent with the enzyme exhibiting pro- and anti-inflammatory properties. Recent reports have linked itaconate production and ACOD1 expression with immunotherapy resistance.

Biological Activity: ERG344 inhibits itaconate production in enzymatic and cell-based assays with an IC50 value of ~200 microM.

ERG344 is manufactured by Ergon Pharmaceuticals in USA and its structure is protected by US Patent 12,060,310. Versions of ERG344 are illegally sold in USA under the name of IRG-IN-1.
